Born on June 30, 1960, Diego Trujillo went to high school at the Gimnasio Moderno School in Bogota, where he occasionally did theater as a hobby. After graduating, he decided to study architecture at the Universidad Piloto de Colombia. He worked as an architect for 12 years, first at an architecture firm, and later as a technical assistant at the company Oxy, in project supervision and on the Cano Limon-Covenas pipeline. He then went on to found an architecture and construction firm in partnership with his former boss.

He saw himself as an entrepreneur, but fate led his friend Felipe Noguera to bring Trujillo to a film set, and he never walked away from it. By chance, he later met director Kepa Amuchastegui, who helped him out in launching his acting career. His acting studies began in 1993 at an improv workshop at the Teatro Libre, and three years later, he took part in a screenwriting workshop with Juana Uribe. That same year, he began his work in theater with the play En carne propia, by director Miguel Torres.

In television, Trujillo has also had a long career, starting in 1992. He has acted in telenovelas including: La maldición del paraíso; El manantial; Mascarada; Perro amor, which won him the Simon Bolivar award and the TV and Telenovelas award for Best Supporting Actor; Porque diablos; El fiscal; Pobre Pablo, which brought him an India Catalina Award for best supporting actor; La costeña y el cachaco; Todos quieren con Marilyn; Los Reyes, one of his best remembered performances, which won him an India Catalina Award for best lead actor in 2005; Infieles anónimos; El capo; La Pola; and ¿Dónde carajos está Umaña?. His most recent role in television was in the telenovela Cuando vivas conmigo, by Caracol.

He has also acted in Colombian TV series such as De pies a cabeza, Tiempos difíciles, and Amas de casa desesperadas. In 2007, he performed in the series Tiempo final, Infieles anónimos, Venenosos, La pócima, Humo, and Cama de hotel, produced by FoxTelecolombia.

In 2008, he ventured into new territory as host of the game show El poder del 10, from the RCN network. In 2014, he played the lead role of Walter Blanco in the series Metástasis, an adaptation of the hit show Breaking Bad, directed by Andres Baiz and co-produced by Sony Pictures Television and Teleset.

His film career began in 2002 with director Taylor Hackford's film Proof of Life, starring Meg Ryan and Russell Crowe, where he was chosen for his excellent English language skills. A year later, he was in Cuando rompen las olas, by director Ricardo Gabrielli. In 2005, he acted in Francisco Norden’s El trato and Harold Trompetero’s Dios los junta y ellos se separan. He worked with director Trompetero again in the feature film Riverside (2009) and, later, in Los oriyinales (2017).

At the screening of Los oriyinales at the Shanghai Film Festival, Diego tried his hand at a new facet of his talent, with his first theatrical monologue Que desgracia tan infinita. He later directed and starred in the monologues Molestia Aparte, Padre rico pobre padre, and most recently, El arte de compartir la tusa: postconflicto. He has acted in other stage plays, including En carne propia, Muelle oeste, La noche árabe, and Cita a ciegas.
